Crospovidone uses in tablets

Crospovidone(PVPP), an insoluble cross-linked polymer, is a high-performance functional excipient in tablet formulations, primarily valued for its disintegration-promoting properties. This excipient possesses a highly developed three-dimensional network structure. Although insoluble in water, it exhibits strong hydrophilicity, initiating swelling within 30 seconds of contact with water, rapidly increasing its volume by 5-10 times.  The resulting expansion pressure directly disrupts the binding forces within the tablet, promoting rapid disintegration into fine particles. Simultaneously, its porous structure facilitates capillary action, accelerating water penetration. This "expansion + capillary" dual mechanism makes it a recognized "super disintegrant," achieving ideal results with only 2%-5% addition in conventional tablets, while the dosage can be adjusted to 5%-8% in special dosage forms such as orally disintegrating tablets.

In various tablet manufacturing processes, crospovidone/PVPP demonstrates excellent adaptability and practicality. Whether in wet granulation, dry granulation, or direct compression processes, it consistently performs effectively.  Its disintegration efficiency can be flexibly controlled through internal addition (mixing with raw materials during granulation), external addition (added during overall granule mixing), or a combination of both, with the latter reducing disintegration time to 5-10 minutes. Its inherent good compressibility and elastic deformation capabilities not only avoid affecting tablet formation during compression but also improve tablet hardness and reduce friability, making it suitable for large-scale production using high-speed tablet presses (speed ≥ 300 tablets/minute).

Crospovidone/PVPP plays a crucial auxiliary role in enhancing drug efficacy, particularly significantly improving the optimization of poorly soluble drugs. The porous channels formed after swelling greatly increase the contact area between the drug and the dissolution medium, increasing the 60-minute dissolution rate of poorly soluble drugs by 20%-30%, directly improving drug bioavailability. Furthermore, it possesses good adsorption and complexation capabilities, forming a stable system with the drug's active ingredients, inhibiting drug inactivation or degradation during the formulation process.  It is also stable in acidic and alkaline environments, making it suitable for various formulations with strict pH requirements. As a non-ionic polymer, it has no incompatibility issues with ionic drug components and can synergistically interact with commonly used excipients such as microcrystalline cellulose and lactose, improving the stability and uniformity of the formulation.

In practical applications, the properties of crospovidone/PVPP make it an ideal excipient choice for various tablets, and it complies with mainstream international pharmacopoeia standards. It can be found in a wide range of products, from common chemical drug tablets such as valsartan amlodipine tablets and rosuvastatin calcium tablets, to health food tablets such as broken-cell Ganoderma lucidum spore powder and coenzyme Q10 tablets. Its low hygroscopicity (moisture absorption rate ≤5% at 25℃/60% RH) prevents caking during storage, maintaining disintegration activity over the long term and reducing formulation storage costs. Furthermore, it does not form a gel even at high concentrations (e.g., 10%), and its particle size is adjustable; smaller particle sizes reduce surface mottling and improve tablet appearance, further expanding its application in various types of tablets.
